Metastasis is one of the major obstacles for breast cancer patients. Limitations of current models demand the development of custom platforms to predict metastatic potential and homing choices of cancer cells. We developed two organ\u2010on\u2010chip platforms, invasion\/chemotaxis (IC\u2010chip) and extravasation (EX\u2010chip) for the quantitative assessment of invasion and extravasation towards specific tissues (lung, liver, breast, and bone), which are the most commonly targeted sites for breast cancer metastasis. Our results show that IC\u2010chip and EX\u2010chip, simulating tissue\u2010specific microenvironments, can distinguish different in vivo metastatic phenotypes of breast cancer cells, in vitro. Furthermore, the systems we developed can be used to assess the effects of anti-metastatic drugs and to model and analyze the molecular mechanisms behind the pre-metastatic niches. IC- and Ex-Chip-based platforms we present provide the groundwork for the development of novel preclinical and clinical tools to predict metastasis risk and understand the underlying molecular mechanisms.<\/p>\n
